The authors isolated a loss-of-function mutation in the egl-1 gene. This mutation prevents most if not all somatic programmed cell deaths. They speculate that Egl-1 activates programmed cell death by binding to and directly inhibiting the activity of Ced-9, conceivably by liberating the cell death activator Ced-4 from a Ced-9/Ced-4-containing protein complex, which is, in turn, free to activate Ced-3, leading to cell death.

Molecular analysis of seven previously identified gain-of-function mutations in egl-1 (causing inappropriate programmed cell death of the HSNs in hermaphrodites; see text), revealed that each was caused by a single base change in a 3′ regulatory region of the gene (a consensus binding site for Tra-1). The authors show that these mutations disrupt the binding of the Tra-1 zinc-finger protein, the terminal global regulator of somatic sexual fate.
