••] demonstrate that PPARγ is required for adipocyte differentiation both in vitro and in vivo. PPARγ null mice completely lacked visible adipose tissues. Mice that are chimeric for wild-type and PPARγ null cells show little or no contribution of null cells to adipose tissue, suggesting that PPARγ is necessary for the development of adipose tissue in vivo. In vitro, the differentiation of embryonic stem cells into adipocytes is completely dependent on PPARγ gene dosage

Puigserver P., Wu Z., Park C.W., Graves R., Wright M., Spiegelman B.M. A cold-inducible coactivator of nuclear receptors linked to adaptive thermogenesis. Cell. 2:1998;829-839. The coactivator PGC-1 potentiates the transcriptional activity of the perioxisome proliferator activated receptor (PPAR)γ on the uncoupling protein (UCP)-1 enhancer. Ectopic expression of this protein in white pre-adipcoytes 3T3-F442A increases the expression of UCP-1 as well as a number of genes involved in respiratory chain function in mitochondria.

An adipogenic cofactor bound by the differentiation domain of PPARgamma
-
PGC-2 binds to the termini of the perioxisome proliferator activated receptor (PPAR)γ at a major domain responsible for adipogenic activity. This coactivator interacts with PPARγ but not other isoforms of PPAR. Ectopic expression of this protein in 3T3-L1 pre-adipocytes enhances adipogenesis
