•] demonstrated the critical role for IL-13 in the manifestation of allergic asthma in the murine models. These studies showed that blockade of IL-13 by a soluble form of the IL-13 receptor (sIL-13Rα2-Fc fusion protein) reversed/ameliorated allergen-induced airway hyper-responsiveness and that administration of IL-13 induced the pathophysiological characteristics of asthma, independent of IgE and eosinophils but dependent on IL-4Rα

Regulation of apoptosis by tyrosine containing domains of IL-4R alpha: Y497 and Y713, but not the STAT6-docking tyrosines, signal protection from apoptosis
-
This report implicated the carboxy-terminal domain (containing Tyr713) in the anti-apoptotic signaling pathway. Interestingly, the Tyr713→Phe mutation in the context of IL-4Rα had no effect on the protection from apoptosis mediated by IL-4. The effect could only be observed in the IL-2Rβ-IL-4Rα chimeric receptor

Cutting edge: Effects of an allergy-associated mutation in the human IL-4R alpha (Q576R) on human IL-4-induced signal transduction
-
This is a follow-up study of reference [48] by a different group. In this study, the mutant (Gln576→Arg) human IL-4Rα gene was stably expressed in a murine myeloid progenitor cell line 32D and in a mouse lymphoma cell line M12.4.1. However the responses generated from the mutant gene were similar to those of the wild-type receptor, implicating that additional alterations in the IL-4 signaling pathway were required for the observed phenotype in cells isolated from Gln576→Arg homozygotes

Ile50Val variant of IL4R alpha upregulates IgE synthesis and associates with atopic asthma
-
This group reported an Ile50 variant of IL-4Rα that associated with atopic asthma. Although the mutation did not significantly affect ligand-binding affinity, human and murine B lymphocyte cell lines expressing the Ile50 variant exhibited higher levels of cell growth, Iε promoter activation and Stat6 activation in response to IL-4 stimulation

Cutting edge: SOCS-1 is a potent inhibitor of IL-4 signal transduction
-
This report implicates SOCS-1 in the negative regulation of IL-4 signal transduction. The expression of SOCS-1 but not SOCS-2 was able to inhibit IL-4-induced activation of a Stat6 reporter in transient transfection. Stable cell lines expressing SOCS-1 showed decreased levels of Jak-1 and Stat6 activation
