Strains were W303 derivatives. The genomic HCT1 open reading frame was tagged with Myc epitopes as described (6). Hct1 is a rare protein. In growing cells, the amount of Myc9-Hct1 was approximately 20-fold lower than that of Cdc23-Myc9 (12). In Fig. 3, A and B, a HA3-HCT1-TRP1 plasmid was integrated at the HCT1 locus.
