CHO-K1 cells stably expressing the sst2, sst3, sst4, and sst5 receptors were subcultured in 12-well plates and treated at 37°C for 30 min with growth medium containing 0.5 mM isobutylmethylxanthine. The medium was replaced with fresh growth medium, with or without 10 μM forskolin and test agents. After incubation for 5 min at 37°C, the medium was removed, and the cells were lysed by freeze-thaw in 0.1 M HCl. The cAMP content of duplicate wells was determined with a radioimmunoassay kit (Amersham). Data obtained from the dose-response curves were analyzed by nonlinear regression with GraphPad Prism, version 2.01 (GraphPad Software, San Diego, CA).
