Spectrophotometric determination of loading capacity was performed by weighing ca. 10 mg dry, Fmoc-functionalized resin into a quartz cuvette. 1.00 mL 20% piperidine in DMF was added to this cuvette and also to a second cuvette as a control. For three minutes, contents of the cuvettes were mixed by gently drawing up and expelling the solution through glass Pasteur pipets (two pipets separately used to prevent cross contamination). Absorbance at 290 nm was measured, and the concentration of released Fmoc was calculated from the difference in absorbances (with baseline corrections) using the following equation. (A290 - A450) - (A290 - A450)control Fmoc concentration =/4950 Finally, loading capacity (mmol/g) was obtained from the number of millimoles released Fmoc (concentration multiplied by 1.00 mL) divided by the amount of weighed resin.
