Typical experimental procedure for Mitsunobu reaction using (N-Boc)-4-hydroxypipecolic acid methyl ester : triphenylphosphine (1.5 mmol) and diethylazodicarboxylate (1.5 mmol) were dissolved in 3 mL ice-cooled distilled THF and stirred for 1 hour. Formic acid (or respectively 2,4-dichlorophenol) (1.5 mmol) was added and the solution stirred for an additional half hour. A solution of the hydroxy acid (1 mmol in 2 mL of THF) was then added dropwise for approximately 1 hour, and the mixture was stirred overnight at r.t.. The desired compound was obtained after evaporation and column chromatography.
