By studying the cellular localization of nine different Cx32 mutants that are associated with X-linked Charcot-Marie-Tooth disease (CMTX) in communication-incompetent mammalian cells, Deschênes et al. show that some of the described CMTX mutations affect both the intracellular trafficking of the mutated connexin polypeptides and the delivery of the connexins to the plasma membrane. This is the first report to suggest that impaired intracellular trafficking of connexin polypeptides could be responsible for some cases of the CMTX syndrome.
