Arterioscler Thromb Vasc Biol 1998; 18:272-276. The authors report that RXR ligand can activate the RXR-PPARα signal transduction pathway in transfected cells as well as in vivo. They also demonstrate that in hypertriglyceridemic db/db mice, RXR and PPARα ligands lower the triglyceride levels, and together these agents exhibit greater efficacy in their triglyceridemia lowering activity. Furthermore, they show that RXR agonists also raise high-density lipoprotein cholesterol levels. These results indicate RXR agonists or their combination with fibrates as novel approaches to treat patients with hypertriglyceridemia and low high-density lipoprotein cholesterol levels.

This paper describes that RXR agonists can function as insulin sensitizers and decrease hyperglycemia, hypertriglyceridemia and hyperinsulinemia in mouse models of type II diabetes. They conclude that the activation of RXR-PPARγ heterodimer with RXR agonists may provide a novel way of treating type II diabetes.
