Kao CM, McPherson M, McDaniel RN, Fu H, Cane DE, Khosla C: Gain of function mutagenesis of the erythromycin polyketide synthase. 2. Engineered biosynthesis of an eight-membered ring tetraketide lactone. J Am Chem Soc 1997, 119:11339-11340. This paper demonstrates two very interesting possibilities: increase in the extent of keto group modification and the formation of an eight-membered lactone ring.

The molecular basis of Celmer's rules: The stereochemistry of the condensation step in chain extension on the erythromycin polyketide synthase
-
Weissman KJ, Timoney M, Bycroft M, Grice P, Hanefeld U, Staunton J, Leadlay PF: The molecular basis of Celmer's rules: the stereochemistry of the condensation step in chain extension on the erythromycin polyketide synthase. Biochemistry 1997, 36:13849-13855. In a mechanistic study it was shown that ketone synthase domains fall into two types: those that epimerise the methyl branching centre and those that do not. This observation is important for attempts to alter the chirality of chain extension.
