This paper describes a general approach to express a fungal type I polyketide synthase (PKS) gene in yeast and E. coli through introduction of the gene for a transferase that adds 4′-phosphopantetheine to the PKS acyl carrier protein domain, to form the fully active PKS. It should be possible to extend this method to bacterial PKSs and other host organisms, such as plants.

This paper reports the first three-dimensional structure of a bacterial polyketide synthase component, the actinorhodin acyl carrier protein (ACP), and shows that it is closely related to the type II fatty acid synthase ACP of E. coli whose structure also was solved by nuclear magnetic resonance methods.

They establish that a putative fatty acid synthase enzyme, malonyl coenzyme A:acyl carrier protein (CoA:ACP) acyltransferase, is required to charge the PKS ACP and that the starter unit is likely to be acetyl-ACP, produced by decarboxylation of malonyl-ACP, and not acetyl CoA itself.

It is an example of process improvement since the fermentation method could supplant the semisynthesis of epirubicin currently used for its manufacture.
