Proc Natl Acad Sci USA 1998, 95:505-509. This paper describes a general approach to express a fungal type I polyketide synthase (PKS) gene in yeast and E. coli through introduction of the gene for a transferase that adds 4′-phosphopantetheine to the PKS acyl carrier protein domain, to form the fully active PKS. It should be possible to extend this method to bacterial PKSs and other host organisms, such as plants.

Nat Biotechnol 1998, 16:69-74. This paper reports the use of deoxysugar biosynthesis genes in combinatorial biosynthesis to make a medically and commercially important antibiotic. It is an example of process improvement since the fermentation method could supplant the semisynthesis of epirubicin currently used for its manufacture.

This paper describes how antibiotic biosynthesis genes cloned from DNA isolated directly from the soil or uncultured bacteria can be used to produce novel polyketides. It is the forerunner of the use of environmental DNA to produce new natural products for drug discovery.
