J Med Chem 1997, in press. Known biphenyl-based selectin inhibitors were redesigned to optimize and augment ligand-receptor interactions. The resulting novel glycomimetics blocked the binding of HL-60 cells bearing sialyl Lewis x to immobilize E-selectin, P-selectin, or L-selectin. Inhibition was potently enhanced when the modified compounds were presented in dimeric or trimeric arrays.

Methodology is presented to generate synthetic structural analogs of naturally occurring saccharide arrays that function as selectin ligands. The ROMP approach (ring-opening metathesis polymerization) was used to synthesize scaffolds presenting multiple copies of sulfated galactose residues that were up to 500-fold more effective at blocking P-selectin-cell interactions than monovalent sialyl Lewis x.

This study describes the heterogeneous adherence repertoire now emerging for H. pylori and demonstrates that sialylated oligosaccharides can effectively prevent bacterial adherence. An especially potent inhibitor of this pathogen is 3′-sialyllactose linked to albumin in a multivalent array.

Libraries of polyacrylates adorned with sialic acid residues and other functionalities were prepared and screened for inhibitory activity against influenza virus binding to erythrocytes. Members of the library shared the same sialic acid content but varied in their presentation of other moieties. Several polymers with particularly potent inhibitory activity were identified. These 'hits' presumably benefitted from fortuitous interactions among their diversity components and undefined viral surface components.

Various synthetic oligosaccharides were evaluated for their ability to neutral-ize anti-αGal IgG. The synthetic free αGal trisaccharide inhibited anti-αGal binding to porcine epithelial cells in the low millimolar range. This result is the first step in developing immunosuppressive agents that will allow pig to human organ transplantation.

Cancer Res 1996, 56:3069-3074. Human cancer cells were enzymatically manipulated in vitro to display the highly immunogenic αGal epitope on their cell surface. High levels of naturally occurring human αGal antibody bound to the modified cells and greatly increased their processing by macrophages. The technique offers a way to stimulate the immune system to prevent and treat cancer.

This work not only generates a potential cancer vaccine but also provides a general strategy for the assembly of molecules in which selected glycoforms can be incorporated at any desired position of the peptide chain.

A combinatorial library of disaccharides attached to solid support was screened for binding activity to the multimeric lectin Bauhinia purpurea. The strongest binding compounds bore little resemblence to the optimal monomeric ligand reported as the 'natural' specificity of the lectin. Even more surprising was the observation that the optimal compounds from the library, when tested as monomers in solution, had a different rank order of binding affinities. This suggests that the context in which carbohydrates are presented can affect the outcome of receptor binding studies.

Of significance is the display on the cell surface of unique functional groups, such as the ketone, that serve as tags for chemoselective conjugation with any molecule containing a hydrazide or aminooxy group.
