The authors report on the kinetics of the interactions between lipopolysaccharide (LPS), LPS-binding protein (LBP) and soluble CD14. LBP interacts simultaneously with both a LPS micelle and soluble CD14 during transfer of an LPS monomer to the soluble CD14. LBP-catalyzed transfer is first order with respect to soluble CD14 and LPS concentration, and the apparent Km values are 1-2 μg/ml for soluble CD14 and 100 μg/ml for LPS. These values are within the range of physiological concentrations for soluble CD14 and LPS.

Immunity 1996, 4:407-414. The role of CD14 in bacterially induced and LPS-induced shock was tested in CD14-deficient mice produced by gene targeting in embryonic stem cells. CD14-deficient mice were found to be highly resistant to shock induced by LPS and bacteria. However, at very high doses of LPS or bacteria, mice died through CD14-independent mechanisms. This report illustrates the dose-dependent role of LPS with regard to CD14, and indicates that, at physiological doses of LPS, CD14 plays a crucial role.

••]). LBP knockout mice were found to be resistant to a low dose challenge of LPS. Furthermore, in these mice peritonitis due to Salmonella typhimurium was lethal whereas heterozygous mice survived. Survival was associated with a decreased number of bacteria. This suggests that LBP could play a benefical role in infection contributing to phagocytosis processes.

The authors have expressed either a full-length CR3 or a mutated form deficient in the cytoplasmic domain in fibroblastic CHO cells. They showed that full-length CR3 was necessary to produce phagocytic signals. In contrast, lipopolysaccharide (LPS) activation did not require the cytoplasmic domain. In fact, LPS activation through CD14 or CR3 of cells occurs without the obligatory participation of a transmembrane receptor.

Tapping RI, Tobias PS: Cellular binding of soluble CD14 requires lipopolysaccharide (LPS) and LPS-binding protein. J Biol Chem 1997, 272:23157-23164. LPS-binding protein (LBP) is traditionally known to function as a catalyst in the transfer of LPS to soluble CD14 (Yu & Wright, 1996 [13]). The data presented in this paper reveals that LBP is actually a physical part of the soluble CD14-containing cell-associated complexes. Cellular binding of soluble CD14 is dependant on LBP and LPS. Once bound, soluble CD14 is internalized suggesting a novel general mechanism by which non myeloid cells clear LPS.
