Selective N-type calcium channel antagonist omega conotoxin MVIIA is neuroprotective against hypoxic neurodegeneration in organotypic hippocampal-slice cultures
-
Pringle AK, Benham CD, Sim L, Kennedy J, Iannotti F, Sundstrom LE. Selective N-type calcium channel antagonist omega conotoxin MVIIA is neuroprotective against hypoxic neurodegeneration in organotypic hippocampal-slice cultures. Stroke 1996; 27:2124-2130. This paper provides a clear demonstration that N-type VSCCs could contribute to ischemic neuronal injury. In the same model, L-type VSCCs do not play a significant role.

Protective effects of extracellular acidosis and blockade of sodium/hydrogen ion exchange during recovery from metabolic inhibition in neuronal tissue culture
-
Vornov JJ, Thomas AG, Jo D. Protective effects of extracellular acidosis and blockade of sodium/hydrogen ion exchange during recovery from metabolic inhibition in neuronal tissue culture. J Neurochem 1996; 67:2379-2389. Our laboratory has focused on mechanisms of ionic injury that occur during recovery from simulated ischemic conditions. These experiments show that rapid restoration of intracellular pH by sodium/hydrogen exchangers could be an important mechanism of reperfusion injury in neurons.

Glutamate receptor-mediated toxicity in optic nerve oligodendrocytes
-
Matute C, Sanchez GM, Martinez ML, Miledi R. Glutamate receptor-mediated toxicity in optic nerve oligodendrocytes. Proc Natl Acad Sci U S A 1997; 94:8830-8835. While glutamate receptors do not play an important role in the short term electrical recovery of the optic nerve, this study suggests that glutamate could injure the oligodendrocytes of white matter under some circumstances.

Mechanisms and effects of intracellular calcium buffering on neuronal survival in organotypic hippocampal cultures exposed to anoxia/aglycemia or to excitotoxins

Abdel-Hamid KM, Tymianski M. Mechanisms and effects of intracellular calcium buffering on neuronal survival in organotypic hippocampal cultures exposed to anoxia/aglycemia or to excitotoxins. J Neurosci 1997; 17:3538-3553. Tymanski and his collaborators have used calcium chelators to investigate precisely how calcium flux and concentration changes contribute to neuronal injury. Here, using organotypic cultures, evidence is presented that presynaptic calcium changes are critical.

Vicious cycle involving Na+ channels, glutamate release, and NMDA receptors mediates delayed neurodegeneration through nitric oxide formation
-
Strijbos PJ, Leach MJ, Garthwaite J. Vicious cycle involving Na+ channels, glutamate release, and NMDA receptors mediates delayed neurodegeneration through nitric oxide formation. J Neurosci 1996; 16:5004-5013. This paper is another important contribution to our understanding of the indirect mechanisms responsible for glutamate release. Using striatal cultures, contributions of nitric oxide and sodium channels were shown to contribute to persistent NMDA receptor mediated injury.
