    • The Drosophila beaten path gene encodes a novel secreted protein that regulates defasciculation at motor axon choice points
    • of special interest. Analysis of a gene mutant in beaten path (beat) flies. Beat is a novel membrane protein required for the selective defasciculation of motor axons at choice points. The beat phenotype is rescued when beat gene is expressed only in motor axons. Mutant motor axons fail to defasciculate and bypass their targets. This phenotype is suppressed by mutations in FasII and comm, two genes encoding cell adhesion molecules. Beat is therefore proposed to be an anti-adhesive protein secreted by motor axons.

    • Presynaptic development at the Drosophila neuromuscular junction: Assembly and localization of presynaptic active zones

This paper describes the extraordinary extent to which presynaptic structures can form at the embryonic neuromuscular junction of Drosophila mutants in which development of postsynaptic somatic muscles is impaired. The analysis of these mutants shows that synapse formation can be separated from the process of target recognition.

    • Prenatal and postnatal requirements of NT-3 for sympathetic neuroblast survival and innervation of specific targets

Thus, other factors that modulate neuronal cAMP-dependent activity may trigger opposite turning behaviours of the growth cone to the same gradient. Such findings may explain how some factors work as both attractive and repulsive agents or how axons can respond similarly to increases or decreases in particular growth factors.

    • The ablation of CR cells, transient neurons in the hippocampus, in organotypic slice cultures prevent the ingrowth of entorhinal but not of commissural afferents. The inhibition of Reelin (an extracellular matrix protein expressed by CR cells) and analysis of reeler (reelin-deficient) mutant mice shows abnormalities in the invasion of entorhinal afferents into specific layers, suggesting that CR cells and Reelin are essential for the formation of layer-specific connections.

Axons were found to branch preferentially on membrane substrates from target layers and showed a clear preference for the target layer when given choices. Heat inactivation of membranes from target layers led to reduced branching. Whereas the same treatment of nontarget layers increased branching. These results suggest that membrane-associated molecules confined to specific layers induce appropriate branches and prevent the formation of inappropriate branches.
