This is the first demonstration of a direct vascular effect of somatostatin in situ, at plasma concentrations that include the physiologic range. Also, it is the first observation that this effect of somatostatin is endothelium-dependent in the rat, and that a peptide can inhibit the influence of NO on resting venous tone. The discussion contains an excellent and thorough summary on the vascular effects of somatostatin.

These in-vitro experiments involve the simultaneous study of venules and their paired arterioles, which are cannulated in series as a bioassay system to detect venular NO release. The findings indicate that the venular endothelium can release a sufficient amount of NO to influence arteriolar tone, which has important implications for the postulated importance of venular arteriolar chemical communication in local blood flow control.

This well designed and rigorous in-vitro study provides important new information on the role of the cyclooxygenase pathway in the response of resistance vessels to reduced oxygen availability. The findings of this study also suggest that altered resistance vessel responsiveness to oxygen is a fundamental vascular alteration that could contribute to the development of hypertension, and that high salt intake, in the absence of hypertension, alters the responsiveness of resistance vessels to endothelium-dependent and -independent vasoactive influences.
