Am J Physiol 1996; 271:H2238-H2245. This is the first demonstration of a direct vascular effect of somatostatin in situ, at plasma concentrations that include the physiologic range. Also, it is the first observation that this effect of somatostatin is endothelium-dependent in the rat, and that a peptide can inhibit the influence of NO on resting venous tone. The discussion contains an excellent and thorough summary on the vascular effects of somatostatin.

The authors use a combination of various inhibitors in endothelium-intact and -denuded segments to characterize the factors released from the venular endothelium in response to different vasoactive agents. This study represents an important contribution to our understanding of endothelial function in the venous microvessels, which have not been widely studied to date.

These in-vitro experiments involve the simultaneous study of venules and their paired arterioles, which are cannulated in series as a bioassay system to detect venular NO release. The findings indicate that the venular endothelium can release a sufficient amount of NO to influence arteriolar tone, which has important implications for the postulated importance of venular arteriolar chemical communication in local blood flow control.

This in-vivo microscopy study is the first to demonstrate that increases in venular shear stress of physiologic magnitude can elicit the release of NO from the venular endothelium, and that this NO diffuses to and dilates nearby arterioles. These findings support the postulated importance of the venular endothelium in local blood flow control.
