Reports the results of DNA analysis of BRCA1 from 263 women with breast cancer seen at several breast cancer clinics that represent a more 'typical' population of women requesting information for inherited breast cancer susceptibility. The data were used to calculate the probability of detecting BRCA1 mutations based on average age of diagnosis in the family, presence of ovarian cancer, both breast and ovarian cancer in a family member, and whether the family is of Ashkenazi Jewish descent. The BRCA1 analysis was performed by conformation-sensitive gel electrophoresis.

This landmark manuscript provides a retrospective analysis of cancer risk, surveillance and risk reduction in persons with an inherited susceptibility to breast and ovarian cancer and provides provisional recommendations for follow-up care for BRCA1 and BRCA2 mutation carriers and their at-risk relatives. This article addresses the need for research protocols to determine the long-term outcome of mutation carriers, efficacy of surveillance methods and prevention strategies to provide sound data for future recommendations for individuals with inherited cancer risk.
