    • The TGF-β family mediator Smad1 is phosphorylated directly and activated functionally by the BMP receptor kinase
    • of special interest. This study identifies conserved carboxy-terminal SSXS motifs as the targets of agonist-induced phosphorylation of pathway-specific SMADs. Evidence is provided that BMP-induced phosphorylation of Smad1 is catalyzed directly by the BMP type I receptor kinase and that it is essential for induced association with Smad4, nuclear accumulation, and transcriptional activity of Smad1.

    • Opposing BMP and EGF signalling pathways converge on the TGF-β family mediator Smad1
    • of outstanding interest. This work identifies a novel mechanism of regulation of Smad1-mediated BMP signaling. It is shown that phosphorylation in the central region of Smad1, catalyzed by Erk MAP-kinases, inhibits nuclear accumulation of Smad1. By this means, receptor tyrosine kinase activating growth factors, such as EGF, can antagonize BMP-induced nuclear activity of Smad1.
