This is an excellent and detailed review of current evidence and thinking as to how oestrogen regulates cyclical gonadotrophin releasing-hormone and gonadotrophin secretion in mammals. Current evidence is strongly in favour of indirect mediation of genomic influences, but still allows for direct actions of oestrogen on the gonadotrophin releasing-hormone neurone.

This study involved elegant utilization of the phenomenon of agonist-induced internalization of G-protein coupled receptors, here measured in situ, to reveal an effect of oestrogen within 30 min on μ-opioid receptors. It is suggested that oestrogen is rapidly stimulating release of endogenous opioids in the specific brain areas examined.

Intracellular recordings reveal that estrogen rapidly reduces the potency of μ-opioid agonists to activate a potassium conductance. The authors suggest that estrogen acts non-genomically through a specific receptor, activating protein kinase A to uncouple the opioid receptor from its potassium channel.

These findings provide a speculative link between two known risk factors in Alzheimer's disease.
