This study used in vivo confocal calcium imaging of intact larval zebrafish to image the activity patterns of a segmentally repeated series of hindbrain neurons during escape behaviors. The results were consistent with the hypothesis that the segmental hindbrain neurons form a functional group. The work is also important as it is the first to image neuronal activity with single-cell resolution in an intact vertebrate.

The authors generated zebrafish larvae with a large fraction of their neurons filled with calcium indicator, which was pressure injected into blastomeres early in development to label the progeny of the blastomeres in the larval fish. This approach allows the generation of neurons filled with the indicator without damage to circuits that may accompany labeling by backfilling via injections into the larval fish.

This elegant work is the first to image neuronal activity at the single-cell level in the intact mammalian brain. The authors used a calcium indicator and two-photon microscopy to study the calcium fluxes in cortical pyramidal cells of rats in response to deflections of their whiskers. The data show a tight relationship between calcium increases and sodium action potentials, but show no evidence for calcium action potentials.

The authors use imaging to study the patterns of elevation of a second messenger (cAMP) induced by neuromodulators applied to neurons in a well-studied motor network. They show that different neuromodulators produce specific patterns of elevation of cAMP among the neurons of the stomato-gastric network in lobsters. These may help to explain the ability of these neuromodulators to produce unique alterations in the motor pattern of the stomatogastric network.

This paper uses imaging with voltage-sensitive dyes to study the influence of movement on the activity of neurons in an invertebrate ganglion. The work shows that blocking gill withdrawal movements does not lead to a dramatic change in the pattern of activity in the abdominal ganglion in response to siphon stimulation, suggesting that sensory feedback from the movement may not exert widespread effects on the neuronal activity in this reflex behavior. This and previous studies from the same group are important as they represent some of the earliest work to image neuronal populations with single-cell resolution during a behavior.
