Spatial and temporal patterns of myosin heavy chain expression in developing rat extraocular muscle

Documentation of the myosin heavy chain expression patterns in maturing extraocular muscle (EOM). These data establish a unique regulatory pattern for embryonic myosin, with both delayed isoform switching and retention of expression into adulthood. In addition, the expression of myosin isoforms traditionally linked to fatigue-resistant fiber types IIA and IIX are surprisingly sparse in EOM and appear late in the maturational sequence. The postnatal onset and limited expression pattern of a myosin that is uniquely observed in EOM (see [7]) is shown to correlate with visuomotor maturation.

Demonstrates that some extraocular muscle (EOM) fibers, particularly those in the orbital layer, do not downregulate an embryonic isoform of neural cell adhesion molecule. These data, together with the continued expression of embryonic myosin [1••,2] and the presence of an EOM-specific myosin [1••,7] in this same muscle layer, establish the particularly unique nature of orbital-layer fiber types.

The authors examined the expression of γ (fetal) and ε (adult) acetylcholine receptor (AChR) subunit protein at neuromuscular junctions of adult rat extraocular muscle (EOM), diaphragm, and limb muscle. They observed restriction of subunit expression to EOM, at endplates of both singly and multiply innervated fiber types: the γ subunit colocalized with E subunit at some, but not all, neuromuscular junctions. They argue that the coexpression of AChRs composed of fetal and adult isoforms at the same neuromuscular junctions might affect the contractile properties and disease susceptibility of EOM.

Although these data are from limb muscle, which degenerates in muscular dystrophy, it is important to understand the primary pathogenic mechanisms so one can figure out how the extraocular muscle (EOM) is protected in this disease [28-30].

Previous studies have established an oxidative stress-based mitochondrial model of aging [43]. The authors extend this model to normal aging muscle. This paper, and others by this group [44-46], establish that this type of aging may be accelerated in extraocular muscle (EOM), thereby potentially explaining the propensity for mitochondrial myopathies in EOM.

This preliminary report examines the role played by the developing visual system in modulation of extraocular muscle (EOM) myosin expression patterns. Strategies were used to alter visual development and the consequences for EOM myosin then evaluated. Alterations in the expression of a myosin unique to the EOMs [7] were of particular interest, as this isoform appears during the postnatal maturation of visuomotor systems [1••]. Both dark rearing and monocular deprivation directly altered eye muscle properties by changing the expression patterns of some of the many myosin isoforms found in EOMs. Notably, mRNA levels for the myosin isoform that is specific for EOMs were reduced dramatically in the fiber types normally expressing this isoform. By contrast, the cellular distribution and levels of the EOM-specific myosin mRNA was not altered in monocularly deprived EOMs. Together, these data suggest that at least some of the unique properties of EOMs might result from the unusual demands that are placed upon these muscles.

Extends the genetic characterization of congenital fibrosis of the extraocular muscles (CFEOM) by this group [66,67], initiating the development of a model for this disease. Reports correlated genetic and morphopathologic analysis of a defined group of patients with CFEOM. These patients exhibit ptosis, external ophthalmoplegia, and downward deviation of the eyes. Absence of the levator palpebrae superioris and reduction of the superior rectus muscle correlate with the loss or reduction in the motoneuron pools innervating these muscle groups. The authors suggest that a disease long thought to result from muscle fibrosis actually may have a neurologic origin. The correlative approach used by the authors is effective in discerning disease mechanisms and should be applied to a broader range of ocular motility disorders.
