    • Genetic susceptibility to death from coronary heart disease in a study of twins
    • This major study shows that death from CAD in people of younger ages is influenced by genetic factors both in female and male. Utilizing data from 26 years of follow-up, the investigators studied the risk of death from coronary heart disease in pairs of 3298 monozygotic and 5964 dizygotic male twins and 4012 monozygotic and 7730 dizygotic female twins. The results illustrated several points. First, a predisposition to death from CAD was present, and decreased with increasing age. The relative hazard of death was 8.1 for monozygotic twins and 3.8 for dizygotic twins when one /Es twin died of CAD before age 55 years. The relative hazard of death was even greater in women, 15 for monozygotic twins and 2.6 for dizygotic twins, when one >Es twin died of CAD before age 65 years

    • This is the first study to implicate the ACE DD genotype as a risk factor for myocardial infarction. ACE I/D polymorphism is a major determinant of the plasma level of ACE, accounting for approximately 50% of its variance. Individuals with the DD genotype have higher plasma ACE level than those with ID or II genotype. Cambien and his colleagues show that DD genotype was more common in patients with Ml than in controls, especially among patients with low body mass index and low plasma level of ApoB

    • Paraoxonase is a HDL associated protein with no known physiologic substrate in humans. However, increasing evidence suggest that paraoxonase plays a significant role in atherosclerosis, most likely through its influence on oxidation of LDL. There is a 10-40 fold variation in plasma paraoxonase level among individuals. There is a genetic basis for such, as large inter-individual variations were shown to be the result of a polymorphism in position 91 of the enzyme where an arginine substitutes for glutamine. Arginine at position 92 confers higher paraoxonase activity than glutamine. This polymorphism has been implicated as a genetic marker for atherosclerosis
