The search for antimicrobial agents effective against bacteria resistant to multiple antibiotics
-
Chopra I, Hodgson J, Metcalf B, Poste G: The search for antimicrobial agents effective against bacteria resistant to multiple antibiotics. Antimicrob Agents Chemother 1997, 41:497-503. A biological approach to the development of novel antibacterial agents is presented, emphasizing attempts to circumvent known resistance mechanisms. Methods to identify targets expressed in vivo or targets specifically involved in infection are discussed.

New directions in antibacterial research
-
Chu DTW, Plattner JJ, Katz L: New directions in antibacterial research. J Med Chem 1996, 39:3853-3874. An excellent perspective is presented from the medicinal chemistry point of view describing target-based antibacterial research. Major groups of compounds directed toward resistant bacteria are presented, with structures, together with possible novel approaches to developing new agents.

Molecular interactions of a semisynthetic glycopeptide antibiotic with D-Alanyl-D-Alanine and D-Alanyl-D-Lactate residues
-
Allen NE, Le Tourneau DL, Hobbs JN: Molecular interactions of a semisynthetic glycopeptide antibiotic with D-Alanyl-D-Alanine and D-Alanyl-D-Lactate residues. Antimicrob Agents Chemother 1997, 41:66-71. Interactions between the novel glycopeptide LY191145 and peptidoglycan dipeptide intermediates are similar to those for vancomycin. The good antibacterial activity of LY191145 against vancomycin-resistant bacteria is thought to be due to strong dimerization of the novel glycopeptide and its more favorable membrane anchoring, leading to increased binding affinities at the bacterial target.

Design, synthesis, and evaluation of 2 β-alkenyl penam sulfone acids as inhibitors of β-lactamases
-
Richter HGF, Angehrn P, Hubschwerlen C, Kania M, Page MGP, Specklin J-L, Winkler FK: Design, synthesis, and evaluation of 2 β-alkenyl penam sulfone acids as inhibitors of β-lactamases. J Med Chem 1996, 39:3712-3722. A series of penam sulfone acids has similar inhibition parameters for class A and class C β-lactamases as the penam sulfone tazobactam, but has different reaction intermediates. Ro 48-1220 enhances the activity of ceftriaxone fourfold to eightfold more effectively than tazobactam against the increasingly important class C cephalosporinase-producing Enterobacteriaceae.

Inhibition of metallo-β-lactamases by a series of mercaptoacetic acid thiol ester derivatives
-
Payne DJ, Bateson JH, Gasson BC, Proctor D, Khushi T, Farmer TH, Tolson DA, Bell D, Skett PW, Marshall AC et al.: Inhibition of metallo-β-lactamases by a series of mercaptoacetic acid thiol ester derivatives. Antimicrob Agents Chemother 1997, 41:135-140. Mercaptoic acid thiol ester analogs were shown to inhibit several subclasses of metallo-β-lactamases. These active-site-directed inhibitors are the first examples of non-β-lactam mechanism-based inhibitors for groups of β-lactamases that have no commercially viable inhibitors.

Antibacterial agents that inhibit lipid a biosynthesis
-
Onishi HR, Pelak BA, Gerckens LS, Silver LL, Kahan FM, Chen M-H, Patchett AA, Galloway SM, Hyland SA, Anderson MS, Raetz CRH: Antibacterial agents that inhibit lipid A biosynthesis. Science 1996, 274:980-989. Lipid A biosynthesis represents an unexploited target for antibacterial research. The most active deactylase inhibitor described was bactericidal and cured mice of a lethal E. coli infection, but was inactive against infections caused by other Enterobacteriaceae, suggesting high bacterial specificity for these inhibitors.
