Antimicrob Agents Chemother 1997, 41:66-71. Interactions between the novel glycopeptide LY191145 and peptidoglycan dipeptide intermediates are similar to those for vancomycin. The good antibacterial activity of LY191145 against vancomycin-resistant bacteria is thought to be due to strong dimerization of the novel glycopeptide and its more favorable membrane anchoring, leading to increased binding affinities at the bacterial target.

A series of penam sulfone acids has similar inhibition parameters for class A and class C β-lactamases as the penam sulfone tazobactam, but has different reaction intermediates. Ro 48-1220 enhances the activity of ceftriaxone fourfold to eightfold more effectively than tazobactam against the increasingly important class C cephalosporinase-producing Enterobacteriaceae.

The most active deactylase inhibitor described was bactericidal and cured mice of a lethal E. coli infection, but was inactive against infections caused by other Enterobacteriaceae, suggesting high bacterial specificity for these inhibitors.

Oxazole isosteres of pseudomonic acid were synthesized with a second mode of action separate from that of isoleucyl-tRNA synthetase inhibition. Both in vitro and in vivo activity of the isosteres were demonstrated.
