Genetic decrease in atrial natriuretic peptide and salt-sensitive hypertension
-
published erratum appears in Science 1995, 267:1753
-
John SWM, Krege JH, Oliver TM, Hagaman JR, Hodgin JB, Pang SC, Flynn TG, Smithies O: Genetic decrease in atrial natriuretic peptide and salt-sensitive hypertension [published erratum appears in Science 1995, 267:1753]. Science 1995, 267:679-681. The ANP gene is disrupted in this work. When ANP-deficient mice were fed high salt (8%) diets, the blood pressures were elevated relative to standard diets, and it was, therefore, suggested that ANP was critical for the handling of dietary sodium by the kidney.

Blood pressure and fluid electrolyte balance in mice with reduced or absent ANP

John SWM, Veress AT, Honrath U, Chong CK, Peng L, Smithies O, Sonnenberg H: Blood pressure and fluid electrolyte balance in mice with reduced or absent ANP. Am J Phsiol 1996, 40:R109-114. This is a second report on the ANP deficient mice. In this paper, anesthetized ANP null mice demonstrated a high blood pressure not affected by dietary salt. There also was no difference between urine flow and sodium excretion in ANP deficient mice and that of wild type mice. The authors concluded that ANP was not essential for chronic regulation of salt balance.

Salt-resistant hypertension in mice lacking the guanylyl cyclase-A receptor for atrial natriuretic peptide
-
Lopez MJ, Wong SKF, Kishimoto I, Dubois S, Mach V, Friesen J, Garbers DL, Beuve A: Salt-resistant hypertension in mice lacking the guanylyl cyclase-A receptor for atrial natriuretic peptide. Nature 1995, 378:65-68. The disruption of the GC-A gene results in chronic elevations of blood pressure which are not affected by high (8%) or low (0.008%) dietary salt. These results suggested that GC-A played a critical role in the chronic regulation of blood pressure but that the receptor was not required for the chronic regulation of salt balance.

The heart communicates with the kidney exclusively through the guanylyl cyclase-A receptor: Acute handling of sodium and water in response to volume expansion
-
Kishimoto I, Dubois S, and Garbers DL: The heart communicates with the kidney exclusively through the guanylyl cyclase-A receptor: acute handling of sodium and water in response to volume expansion. Proc Natl Acad Sci U S A 1996, 93:6215-6219. In anesthetized GC-A deficient mice, the expansion of plasma volume by intravenous infusion caused release of atrial granules but natriuresis or diuresis was impaired. These studies suggested that GC-A is a pivotal kidney receptor for natriuretic or diuretic factors, or both, released from the heart.

-
Huang PL, Huang Z, Mashimo H, Bloch KD, Moskowitz MA, Bevan JA, Fishman MC: Hypertension in mice lacking the gene for endothelial nitric oxide synthase. Nature 1995, 377:239-242. In this study where the eNOS gene was disrupted, acetylcholine-induced relaxation of aortic rings was absent in eNOS deficient mice. The blood pressure of these mice are higher than those of wild type. The results suggest a major role for the soluble guanylyl cyclase in chronic and acute blood pressure regulation.
