    • Neuregulins are concentrated at nerve-muscle synapses and active ACh-receptor gene expression
    • of special interest. In this study, immunohistochemistry is used to demonstrate that NRGs are localized to synaptic sites of skeletal muscle. Furthermore, recombinant NRG stimulates tyrosine phosphorylation of ErbB2 and ErbB3 in C2 myotube cells and upregulates expression of a reporter construct containing the acetylcholine receptor δ-subunit promoter element. Together, these data provide evidence that NRG signaling mediated by ErbB2 and ErbB3 stimulates acetylcholine receptor expression at synaptic sites in skeletal muscle.

    • ARIA is concentrated in nerve terminals at neuromuscular junctions and at other synapses
    • of special interest. In this report, antibodies are used to demonstrate that NRG is found in motor neuron cell bodies and axons, indicating that NRG protein could be transported from the cell bodies in the spinal cord into the nerve terminal. In addition, it is shown that NRGs are localized to synapses in the CNS (e.g. the synapse-rich molecular layer of the cerebellum). The most interesting finding in this paper is that antibodies directed against specific NRG isoforms react with functionally distinct classes of neurons.

NRG protein was found to be present not only at synaptic sites, but also in extrasynaptic regions and in the cytoplasm of the muscle fiber. • of outstanding interest. An activity present in DRG-conditioned medium prevents rat Schwann cell precursors isolated from embryonic nerve from undergoing apoptotic cell death. Axonal interactions regulate Schwann cell apoptosis in developing peripheral nerve: neuregulin receptors and the role of neuregulins. of special interest J Neurosci. 16:1996;6107-6118 Programmed cell death is reported in Schwann cells of early postnatal sciatic nerve which is upregulated by axotomy and can be inhibited by NRG. In addition, phosphorylated forms of ErbB2 and ErbB4 are found to be expressed in these cells.

    • Schwann cell apoptosis at developing neuromuscular junctions is regulated by glial growth factor
    • of outstanding interest. It is reported that denervation of neonatal, in contrast to adult, sciatic nerve induces apoptosis of Schwann cells at neuromuscular junctions, which can be prevented by the in vivo infusion of NRG, implicating NRG as a axonal-derived survival factor for neonatal terminal Schwann cells.
