Global analysis of a macromolecular interaction measured on BIAcore®
-
22. Roden LD, Myszka DG: Global analysis of a macromolecular interaction measured on BIAcore®. Biochem Biophys Res Commun 1996, 225:1073-1077. This paper demonstrates that an antigen-antibody interaction measured on BIAcore® 2000 could be described by a simple bimolecular interaction mechanism. Therefore, effects related to the sensor surface, such as mass transport and the dextran matrix, did not complicate the observed binding responses.

-

-
23. Karlsson R, Falt A: Experimental design for improved kinetic analysis of protein protein interactions with surface plasmon resonance biosensors. J Immunol Methods 1996, in press. This is an excellent paper that presents the experimental conditions required to performed a detailed kinetic analysis. Using global analysis, the authors demonstrated that one antigen-antibody reaction was described by a simple interaction model, but a second reaction required additional sensor experiments to discriminate between complex binding models. They also showed experimentally that no binding artifacts were associated with the dextran matrix.
