Polymer solutions made from DMPA were provided for both DNA sieving and inhibition of electro-osmosis. They also discuss the use of a copolymer that self-assembles into extended network structures with a well-defined pore size permitting DNASCE. This polymer, poly(ethylene glycols) end-capped with micelle-forming fluorocarbon tails, also formed flowable aqueous networks.

Electrophoresis 1996, 17:1103-1109. This paper reports measurements characterizing the dependence of mesh size versus concentration of well-defined semidilute polyacrylamide solutions. The mesh size depended only on the concentration and was independent of the average molecular weight of the polyacrylamide. Electrophoretic mobilities of DNA moving through semidilute polyacrylamide solutions depended almost entirely on concentration and not on polyacrylamide molecular weight or polydispersity. Peaks, however, were less broadened when the polyacrylamide solutions were of higher molecular weight.

They reaffirm prior work in that diffusion and, to a lesser extent, electrokinetic sample injection are the primary band broadening mechanisms in DNASCE . They also show, however, that the standard approach to estimating the diffusion coefficients (i.e. the Einstein relation) is fundamentally wrong and invalid under most DNASCE conditions.

Electrophoresis 1996, in press. This experimental work clearly illustrates that elevated temperature may substantially enhance the resolution in DNASCE. For their 3% polyacrylamide solution and specific set of run conditions, the authors found that an optimum temperature of 55°C provided the best resolution of sequencing fragments up to 848 bases with optimized base-calling software.

The electrophoretic mobilities of DNA restriction fragments were carefully studied for coated versus uncoated capillaries and found to be identical.

This paper presents measurements of the surface roughness of standard capillaries, from Polymicro Technologies (Phoenix, Arizona), using an atomic force microscope. The inner capillary surface displayed remarkably smooth features with an average surface roughness of 2.8-6.7 A. It was concluded that because the surface roughness is much smaller than the diffuse double layer, it cannot possibly influence peak shape and contribute to band broadening in capillary electrophoresis.

These polymer solutions have the interesting property that when heated the viscosity dramatically drops, due to a phase transition, allowing easy capillary loading.

This paper reports the use of a copolymer that self-assembles into extended network structures with a well-defined pore size, permitting DNASCE. These polymers, poly(ethylene glycols) end-capped with micelle-forming fluorocarbon tails, formed flowable aqueous networks that achieved read lengths near 350 bases with DNASCE.
