In this paper the concerted interaction of enzymatic and nonenzymatic lipid peroxidation was studied in vitro. The reconstituted system used may be regarded as a suitable model system reflecting the oxidative events in the developing lesion. A major finding of this study was the fact that 15-LOX catalysed LDL oxidation proceeds in the presence of lipophilic antioxidants. This result may be important for strategies aimed at designing antioxidants for treatment of atherosclerosis.

J Biol Chem 1995, 270:5191-5197. This paper provides, for the first time, direct evidence that overexpression of intracellular 15-LOX may lead to an increase in LDL oxidation. These data were obtained with genetically manipulated cells (15-LOX transfectants) in vitro. It remains to be established whether similar effects are also observable with natural cells expressing the 15-LOX such as interteukin-4-treated monocytes.

Sun D, Funk C: Disruption of 12/15-lipoxygenase expression in peritoneal macrophages. J Biol Chem 1996, 271:24055-24062. The disruption of the 12/15-LOX gene in mice is reported in this paper. Using unspecific markers for lipid peroxidation (thiobarbituric acid-reactive substances, isoprostanes), the authors showed that zymosan stimulated macrophages of these animals have a reduced LDL oxidizing capacity compared with macrophages from normal mice. The major methodological breakthrough associated with this paper is that these knockout mice may be used in the future for cross-breeding studies with other mouse atherosclerosis models such as apolipopotein E knockout mice or LDL receptor deficient mice, to examine the role of 12/15-LOX in atherogenesis in vivo.

FASEB J 1995, 9:1623-1631. Here the creation and characterization of transgenic rabbits that overexpress the 15-LOX selectively in monocyte/macrophages is reported. In order to achieve macrophage-specific expression, a chicken lysozyme promotor was used. A drawback of this promotor is that the regulation of the expression of the transgenic 15-LOX will be different from that of the normal enzyme. On the other hand, the advantage of using this promotor is that the transgene is selectively expressed in the macrophages, in a cell in which the enzyme is induced during atherosclerosis. Thus, the role of macrophage-specific 15-LOX expression can be examined.
