Complement regulation in the rat glomerulus: Cryy and CD59 regulate complement in glomerular mesangial and endothelial cells
-
Quigg RJ, Morgan BP, Holers VM, Adler S, Sneed AR III, Lo CF: Complement regulation in the rat glomerulus: Cryy and CD59 regulate complement in glomerular mesangial and endothelial cells. Kidney Int 1995, 48:412-421. This paper investigated the relative contribution of rat Crry (the rat membrane cofactor protein/decay accelerating factor analog) and CD59 in protecting endothelial cells and masangial cells for complement mediated cytotoxicity. They showed that Cryy played a more prominent role in controlling alternative pathway mediated events.

Role of the complement membrane attack complex (C5b-9) in mediating experimental mesangioproliferative glomerulonephritis

Brandt J, Pippin J, Schulze M, Hansch GM, Alpers CE, Johnson RJ, Gorson K, Couser WG: Role of the complement membrane attack complex (C5b-9) in mediating experimental mesangioproliferative glomerulonephritis. Kidney Int 1996, 49:335-343. These authors present evidence that suggest that C5b-9 (MAC) plays a significant role in causing the damage induced by complement in the antithymocyte serum model. They used the PVG C6-rat model.

The effects of soluble recombinant complement receptor 1 on complement-mediated experimental glomerulonephritis

Couser WG, Johnson RJ, Young BA, Yeh CG, Toth CA, Rudolph AR: The effects of soluble recombinant complement receptor 1 on complement-mediated experimental glomerulonephritis. J Am Soc Nephrol 1995, 5:1888-1894. This work showed that sCR1 can significantly mitigate complementmediated tissue damage and renal pathology in several model systems of GN. These results may have therapeutic implications since sCR1 is now in clinical trials to evaluate its anti-inflammatory potential.

This study demonstrated a relationaship between clinical outcome and urinary SC5b-9 levels. Elevated SC5b-9 levels were correlated positively with poor prognosis and thus may be a dynamic marker of ongoing tissue injury and offer a means of monitoring the effectiveness of immunosupressive therapy.

Complement inhibition with an anti-C5 monoclonal antibody prevents acute cardiac tissue injury in an ex vivo model of pig-to-human xenotransplantation
-
Kroshus TJ, Rollins SA, Dalmasso AP, Elliott EA, Matis LA, Squinto SP, Bolman RM III: Complement inhibition with an anti-C5 monoclonal antibody prevents acute cardiac tissue injury in an ex vivo model of pig-to-human xenotransplantation. Transplantation 1995, 60:1194-1202. These authors described an anti-C5 monoclonal antibody that effectively inhibits C5 conversion and thus limited HAR in this model system. As with sCR1, this monoclonal may have therapeutic potential in the renal setting.

Transfected CD59 protects mesangial cells from injury induced by antibody and complement
-
Nangaku M, Meek RL, Pippin J, Gordon KL, Morgan BP, Johnson RJ, Couser WG: Transfected CD59 protects mesangial cells from injury induced by antibody and complement Kidney Int 1996, 50:257-266. This paper demonstrated the potential effectiveness of transfecting CD59 into mesangial cells, which overexpressed the protein and protected the cells from complement attack. This work represents the application of gene therapy technology to treat renal disease and thus may be the first step in a long path to achieving clinically useful therapy with this technology.
