Any background signal (fluorescence or scattered light) from the buffer should be subtracted, if it is significant. Likewise, if the ligand titrant contributes to the signal (when the fluorescence from the macromolecule is being observed), then these contributions should be subtracted. The observed fluorescence signal of the sample either can be measured at a single emission wavelength or can be integrated over part or all of the emission envelope. If measurements are made at one emission wavelength, then it should be realized that the enhancement factors may depend on the selected wavelength, if there is a red or blue shift in fluorescence on binding. In either case, it is usually not necessary to correct the signals for any wavelength dependence of the photomultiplier response, since only relative signals are needed for the purpose of studying ligand binding.
