Wen W. Meinkoth JL, Tsien RY, Taylor SS: Identification of a signal for rapid export of proteins from the nucleus. Cell 1995, 82:463-473. This report and [9**] describe the first examples of leucine-rich nuclear export signals. Short peptides containing the export signal under investigation were conjugated to bovine serum albumin or glutathione S-transferase and microinjected into the nucleus. These hydrophobic peptides were capable of targeting the export of the conjugate to the cytoplasm. Mutation of hydrophobic residues within these sequences prevented cytoplasmic targeting. This type of experiment has become the hallmark for demonstrating the existence of a transferable nuclear export signal.

Mutations in elF-5A result in a block in Rev-mediated nuclear export and Rev trans-activation and inhibit viral replication.

This study provides compelling evidence for a link between the protein import machinery and the directional export of U snRNAs. A complex of the CBC, capped RNA and importin α is identified. This complex is disrupted by importin β providing a mechanism for the release of RNA after export.
