This report and [9**] describe the first examples of leucine-rich nuclear export signals. Short peptides containing the export signal under investigation were conjugated to bovine serum albumin or glutathione S-transferase and microinjected into the nucleus. These hydrophobic peptides were capable of targeting the export of the conjugate to the cytoplasm. Mutation of hydrophobic residues within these sequences prevented cytoplasmic targeting. This type of experiment has become the hallmark for demonstrating the existence of a transferable nuclear export signal.
