Positional cloning moves from perditional to traditional
-
Collins FS: Positional cloning moves from perditional to traditional. Nature Genet 1995, 9:347-350. An interesting article presenting an historical overview of positional cloning. Ten years earlier, the thought of mapping disease genes by chromosomal location was incomprehensible. With the development of highly informative genetic markers, however, more than 40 disease genes had been mapped by means of positional cloning through 1995. Future speculation is made regarding the impact of the Human Genome Project through the year 2010 and the identification of disease genes.

Shore EM, Kaplan FS: Tutorial: molecular biology for the clinician, part II. Tools of molecular biology. Clin Orthop Rel Res 1995, 320:247-278. This review is the second of a comprehensive three-part series discussing molecular biology techniques. These reviews are written for the clinician and do not require a broad molecular biology background. Part 1 covers pertinent background information regarding the basics of molecular biology (DNA structure, transcription, and translation and gene regulation). Part 2 reviews the techniques used in positional cloning (cloning vectors, DNA sequencing, Southern blots, PCR, and genomic libraries).
