Homozygous deficient apoC-III mice have no detectable apoC-III protein or mRNA in liver or intestine, whereas apoA-I and A-IV are detected in the liver but reduced in the intestine. This suggests that the intestinal specific element is common to all three genes. Fasting TG levels are 70% of normal and there is no increase in TG after a fat meal.

J Clin Invest 1995, 96:2601-2605. In animals and cell lines apoC-III is downregulated by insulin and a negative IRE has been identified previously in the apoC-III promoter. Using CAT as a reporter gene, variation at any of five variant sites in the apoC-III promoter resulted in loss of response to insulin downregulation. The IRE was mapped to sites -455 and -482; however, only the -455 polymorphic site showed binding to a nuclear protein and this might be the critical site for insulin control.

Circulation 1996, 94:1-274, 1591. Endoscopic biopsies from 18 healthy individuals were used to study apoA-I and apoA-IV synthetic rates and mRNA levels; these were highly correlated. When these individuals were haplotyped for the five APOC-III promoter polymorphisms, the two resulting haplotypes were significantly associated with apoA-I and apoA-IV synthetic rates. The intestinal element for all three genes of the cluster might lie within this region of the APOC-III promoter.

The intestinal expression enhancer of the human APOA-I C-III A-IV cluster was mapped to a region at -780 to -520 within the APOC-III promoter with cis-acting suppressors lying outside the region -1300 to - 200. Two specific 21-mers lying between - 745 to -725 and -700 to -680 bind nuclear proteins from intestinally derived cells. The 260 bp fragment was conserved in mouse apoC-III and is the first in-vivo system defining tissue-specific expression.

Arterioscler Thromb Vasc Biol 1996, 16:941-947. The association of a number of quantitative traits and the five apoC-III promoter polymorphisms and Sstl site was examined. The Sstl site alone was associated with hypertriglyceridaemia but not apoC-III level, carotid thickness or raised cholesterol. The 5′ variable sites had no effect on baseline lipid levels. Thus in the haplotype analysis the significant association with hypertriglyceridaemia was solely attributable to variation at the Sstl site.

The Hutterites represent an isolated community, very useful in genetic studies. Association with variable sites in nine candidate genes and plasma lipid levels was studied. Modest but statistically significant variation in lipid levels could be explained by variation in these candidate genes. Information is lost by analysing males and females together, thus gender-specific differences are not observed.

A statistically significant difference was seen with both polymorphisms. This confirms frequency differences seen in Caucasian populations.
