The achondroplasia mutation, as well as several other possible substitutions at the same or an adjacent site, shows constitutive activation of the receptor kinase. There is a correlation between the capacity to participate in hydrogen bond formation and kinase activation.

This paper shows that the achondroplasia and TD mutations result in constitutive activation of the FGFR3 receptor kinase. They also demonstrate that the receptor kinase activity may or may not retain ligand responsiveness depending on the mutation; there is an approximate correlation between the level of kinase activation and the severity of the phenotype.

    • Fibroblast growth factor receptor 3 is a negative regulator of bone growth

These two papers [31•,32•] show that mice deficient for FGFR3 exhibit prolonged endochondral bone growth which causes skeletal abnormalities such as disproportionately extended limbs. These findings together with histological evidence demonstrate a role for FGFR3 as a negative regulator of endochondral ossification. This work provides a conceptual framework for understanding the consequences of inappropriate FGFR3 activation found in various chondrodysplasias.

This is the first report of constitutive receptor activation caused by a mutation found in a Crouzon syndrome patient. This paper describes the effect of the FGFR2 mutation in a mesoderm induction assay using Xenopus animal caps. Induction of mesoderm in the absence of ligand indicates that the mutation causes a constitutive activation of the receptor.

Proc Natl Acad Sci USA 1996, 93:7894-7899. A number of different mutations found in Crouzon patient genomic DNAs were tested using FGFR-Neu receptor chimeras. Introduction of chimeric receptors containing the mutant extracellular domains of FGFR2 linked to the kinase domain of the Neu receptor resulted in a ligand-independent activation of the Neu receptor kinase and transformation of the recipient assay cells. All the mutations tested caused the activation of the receptor kinase, suggesting a common mechanism for this and similar craniosynostosis syndromes.
