Neurology 1996, 47:1247-125. A report by the Dana consortium on therapy for HIV dementia and related cognitive disorders confirmed the American Academy of Neurology's algorithm and found it rare to have both cognitive and functional impairments without associated neurologic and behavioral deficiencies. Dementia was found to be an independent predictor of physical outcome in affected patients.

Science 1996, 274:1917-1921. The levels of inducible nitric oxide synthase in severe ADC coincided with increased expression of the HIV-1 coat protein gp41 in affected brain tissue. Inducible nitric oxide synthase was induced by gp41 in primary cultures of mixed rat neuronal and glial cells. The data suggest that gp41-induced nitric oxide formation may contribute to the severe cognitive dysfunction associated with HIV-1 infection.

This model of HIV-1 encephalitis utilizes SCID mice lacking functional B and T-cells due to a defect in T-cell receptor and immunoglobulin variable chain rearrangement. This allows the animals to accept human monocytes without rejection. The model demonstrates that human monocytes can synthesize neurotoxins that cross species (human to mouse) and can reproduce key aspects of HIV encephalitis in the animals.
