The incorporated cyclobutane uracil dimer (U=U) contains an isosteric formacetal linker instead of the bridging phosphate (p). This allows synthesis of the lesion in large quantities as required for systematic sequence- and distance-dependent repair studies. The formacetal linkage is alsoan isosteric phosphodiester substitute (X-ray crystallography). The formacetal-linked dimer is a substrate for photolyases from Neurospora crassa and Anacystis nidulans (We thank Prof. Dr. A. P. M. Eker for a generous gift of both enzymes.) Synthesis of this model compound as well as its X-ray structure, incorporation into oligonucleotides, and repairability by the DNA-repair enzymes will be reported. Laser-desorption mass spectra (negative ions detected): calcd for 11: 3548.6, found: 3549; calcd for 12: 3548.6, found: 3548.
