The authors treated mice with SCF prior to and/or after total body irradiation. Eighty percent of mice who received SCF prior to irradiation had long-term survival whereas mice who did not receive SCF or received SCF after irradiation all died. One hundred percent of mice who were administered SCF both prior to and after irradiation had long-term survival. Bone marrow cellularity decreased to approximately 14% of baseline levels within 3 days of irradiation but increased rapidly in mice who received SCF before and after irradiation. In addition, both mature and primitive marrow progenitors that decreased to less than 1% of baseline levels on day 1 postirradiation increased markedly by 6 days after irradiation in SCF-treated animals.

Stem cell factor did not stimulate the proliferation of tumor cell lines in culture. However, SCF did increase colony formation by cell lines that expressed the receptor for SCF. When these cells were tranplanted into nude mice, growth of tumor cells was not enhanced by administration of SCF. Finally, SCF did not protect cell lines from 2 GY of irradiation, irrespective of whether the cell lines expressed the receptor for SCF, or protect xenographs of a cutaneous T-cell lymphoma in nude mice.

This report is the first definitive study documenting that tumor cells in the graft can contribute to relapse after autologous marrow transplantation. The authors marked marrow cells (and occult contaminating tumor cells) with the gene for neomycin resistance and followed patients until relapse. Malignant cells from two patients who relapsed after autologous marrow transplant contained the neomycin-resistance gene.

    • Ex vivo expansion of CD34+ peripheral blood progenitor cells: Implications for the expansion of contaminating epithelial tumor cells
    • + progenitors contaminated with epithelial tumor cells conferred a proliferative advantage to the hematopoietic progenitors. The limitation of this paper is that the tumor cells cultured in these experiments were from established cell lines. How tumor cells that endogenously contaminate hematopoietic progenitors behave in expansion cultures is uncertain.
