This study is designed to evaluate the Na-K-Cl cotransporter system in TM cells. TM cells exhibited cotransport activity, which was inhibited by bumetanide and ethacrynic acid, as well as being modulated by a number of hormones and neurotransmitters. Stimulation of the cotransporter increases cell volume, whereas inhibition decreases cell volume. Hypertonicity-induced cell shrinkage increases TM cell monolayer permeability.

Analysis of trabecular meshwork cultures shows that only part of this effect can be explained by i ndomethacin-induced decrease of the stimulation of cAMP production by epinephrine.
