This paper describes chimeric receptor experiments showing that achondroplasia (transmembrane) mutation alone can activate a transmembrane receptor, probably by promoting ligand-independent dimerization. It offers a mechanism to explain how the achondroplasia mutation constitutively activates FGFR3.
