Wild-type transferrin receptor and several deletion mutants of this receptor lacking portions of the cytoplasmic tail were expressed in cultured hippocampal neurons using defective herpes virus. Mutants lacking cytoplasmic amino acids 7-10, 11-14 or 19-28 (which includes the YTRF [single-letter amino acid code] internalization signal beginning at residue 20) are targeted to both axons and dendrites. These regions do not overlap with portions of the cytoplasmic tail that have been shown to be important for basolateral targeting of the transferrin receptor in MDCK cells [10].

Axonal delivery of β-APP requires the following two regions in the ectodomain: a membrane-proximal segment within the β-amyloid domain, and a more distal region that includes putative N-glycosylation sites.

These investigators show that two previously identified adaptor complex subunit homologs, p47 and β-NAP [24,25], can be co-immunoprecipitated in an adaptor-like heterotetrameric complex. The complex is recruited to Golgi membranes in the presence of GTP-γS, suggesting that it functions in a vesicle coat. This complex does not co-purify with clathrin-coated vesicles, and appears by immunogold electron microscopy to be associated with a novel vesicle coat that is thinner than clathrin coats.

These authors cloned the cDNA encoding a novel homolog of the adaptor complex a chain, and showed that it is present in a ubiquitously expressed adaptor-like complex (termed AP3) with a subunit that cross-reacts with antibodies to the neuron-specific adaptor complex subunit, β-NAP. In yeast two-hybrid studies, the putative μ chain of this ubiquitous complex, p47A, binds to the YQRL (single-letter amino acid code) sorting motif from the trans-Golgi network protein TGN38. The neuron-specific homolog of the μ chain, p47B, interacts with this motif with significantly lower affinity.

Transgenic mice expressing a constitutively active mutant of human Rac1 under the control of the Purkinje-cell-specific L7 promoter perform poorly in tests of cerebellar function, consistent with a defect in cerebellar circuitry. Purkinje cells develop normal dendritic arbors but with numerous, unusually small spines; axonal terminal fields in the deep cerebellar nuclei are reduced.
