The authors study the apoptotic effect of ultraviolate B on HeLa cells and cultured human vascular endothelial cells and document the extracellular display of phosphatidylserine with resultant procoagulant activity. They propose that this exposure of anionic phospholipids may also induce the antiphospholipid antibodies occasionally seen in LE.

J Immunol 1996, 156:4059-4066. Rabbits were immunized with 15- to 18-mer peptides from the sequence of the 60-kD Ro antigen. These peptides were known to be either bound or not bound by human anti-Ro sera. Animals produced antibodies to the immunizing peptides as well as autoimmune responses to serial 8-mer peptides from the entire 60-kD Ro antigen. A proportion of animals developed antibody responses to the La antigen as well. In each case there was evidence of epitope spreading, revealing that immunization with a self-peptide can ultimately result in the processing and successful presentation of a self-protein to T cells.

Lupus-prone MRL/lpr mice were rendered deficient in either CD40 ligand, αβ T cells, or both by mating the MRL/lpr mice with CD40 ligand-deficient mice and T-cell receptor α chain-deficient mice. The autoimmune phenotypes were then analyzed. Skin disease phenotype did not strictly correlate with renal disease, suggesting different pathogenetic mechanisms: CD40 ligand-negative, αβ T-cell receptor-positive mice developed low levels of autoantibodies and mild renal disease yet had unchanged cutaneous disease. CD40 ligand-negative, αβ T-cell receptor-negative animals developed higher levels of anti-double-stranded DNA autoantibodies and developed renal disease yet failed to develop skin disease. Persistence of skin disease in animals singly deficient in αβ T cells or CD40 ligand argues against a solely αβ T cell-dependent autoantibody mechanism. The absence of disease in doubly deficient mice was correlated to the absence of complement-fixing IgG2a isotype.

In situ expression of the B7 and CD28 receptor families was examined in active skin lesions as well as in nonlesional skin of patients with SLE, SCLE, and CCLE and healthy controls using immunocytochemistry and RT-PCR. B7-1 (CD80) and B7-2 (CD86) were found to be expressed on dermal and (minimally) on epidermal antigen-presenting cells and T cells. This compartmentalization was confirmed by comparative RT-PCR of skin biopsy specimens and suction blisters. These cells were able to bind CTLA-4 immunoglobulin in situ. CD28 was expressed by most T cells infiltrating either the dermis or epidermis. Nonlesional skin in subjects and normal controls was negative for these markers.

A conalbumin-reactive T-helper 2 T-cell clone was induced to overexpress LFA-1 by transfection and was subsequently shown to lyse autologous macrophages and to induce autoimmunity in congenic mice in the form of anti-DNA responses, immune complex kidney disease, and pulmonary alveolitis. Treatment of the parent clone with procainamide and ensuing DNA hypomethylation resulted in similar LFA-1-dependent autoimmunity. The authors thus propose that DNA hypomethylation-induced LFA-1 overexpression may be sufficient to break tolerance, presumably by enhancing cross-reactive specificities of T cells.

To study the local cytokine profile in cutaneous LE, RT-PCR was performed on lesional skin from 19 patients with cutaneous LE (eight with SLE, two with SCLE, and nine with CCLE) and four healthy controls. Analysis of interleukin-2, -4, -5, and -10 and interferon gamma mRNA expression was performed. Interleukin-10 and interferon gamma were found in the majority of specimens, including normal controls. Interleukin-5 was found 15 of 19 LE specimens and in none of the controls. The pattern of cytokine mRNA expression did not vary with different subtypes of cutaneous LE. These results indicate a mixed cytokine pattern possibly favoring cell adhesion via interferon gamma-mediated increases in ICAM-1 expression and a T-helper 2 response with significant interleukin-5 production and detectable interleukin-10 production.

J Immunol 1996, 156:3043-3050. The lupus-prone mouse strain NZW shows a reduced production of TNF-α as does a subset of patients with SLE. This is associated with a polymorphism in a simple tandem repeat sequence within the promoter region of the gene. Using a luciferase reporter system and TNF-α promoter constructs from various mouse strains, the authors show that mutations in the 3′ untranslated region of the TNF-α gene play an important role in the downregulation of TNF-α biosynthesis. Interestingly, although luciferase levels varied, there were no significant differences in luciferase mRNA levels, paralleling a discord in TNF-α protein and mRNA levels seen in these different genetic backgrounds, suggesting that the alteration in the 3′-untranslated region affects primarily posttranslational regulation of protein synthesis.
