Alterations in the hypothalamic paraventricular nucleus and its oxytocin neurons (putative satiety cells) in Prader-Willi syndrome: A study of five cases
-
Swaab DF, Purba JS, Hoffman MA: Alterations in the hypothalamic paraventricular nucleus and its oxytocin neurons (putative satiety cells) in Prader-Willi syndrome: a study of five cases. J Clin Endocrinol Metabol 1995, 80:573-579. Interesting study that reports some of the first autopsy data on the immunochemistry of brain tissue in persons with PWS. The paper is weakened by incomplete proof of the presence of PWS in study patients as well as by a poor control group. Nevertheless, conclusions point in an interesting and perhaps an important direction for future research.

Genomic imprinting in the Prader-Willi and Angelman syndromes
-
in press
-
Glenn CC, Driscoll DJ, Yang TP, Nicholls RD: Genomic imprinting in the Prader-Willi and Angelman syndromes. Molec Hum Reprod, in press. This review describes in detail the various genetic mechanisms that result in PWS and Angelman syndrome and the genes discovered in patients with PWS. It also describes the imprinting center, a novel genetic element that controls the initial resetting of the parental imprint in the germline for all imprinted gene expression over a 1.5-2.5 Mb region within chromosome 15q11-q13.

Inherited microdeletions in the Angelman and Prader-Willi syndromes define an imprinting center on human chromosome 15
-
Buiting K, Saitoh S, Gross S, Dittruch B, Schwartz S, Nicholls RD, Horsthemke B: Inherited microdeletions in the Angelman and Prader-Willi syndromes define an imprinting center on human chromosome 15. Nat Genet 1995, 9:395-400. The authors present data on three persons with PWS and two persons with Angelman syndrome who have inherited microdeletions within chromosome 15q11-q13. They propose that 15q11-q13 contains a single imprinting center rather than separate paternal and maternal imprinting control elements.

Minimal definition of the imprinting center and fixation of a chromosome 15q11-q13 epigenotype by imprinting mutations

Saitoh S, Buiting K, Rogan PK, Buxton JL, Driscoll DJ, Arnemann J, Konig R, Malcolm S, Horsthemke B, Nicholls RD: Minimal definition of the imprinting center and fixation of a chromosome 15q11-q13 epigenotype by imprinting mutations. Proc Natl Acad Sci U S A 1996, 93:7811-7815. This paper describes four new microdeletions of the imprinting center. The data presented here strongly support a model in which the paternal chromosome in PWS can carry an ancestral maternal epigenotype and the maternal chromosome of persons with Angelman syndrome may carry an ancestral paternal epigenotype.

Toth-Fejel S, Olson S, Gunter K, Quan F, Wolford J, Popovich BW, Magenis RE: The impact of imprinting: Prader-Willi syndrome resulting from chromosome translocation, recombination and nondisjunction. Am J Hum Genet 1996, 58:1008-1016. A report of two cases of PWS with UPD in which a meiosis I nondisjunction error involving an altered chromosome 15 produced by both a translocation event and recombination was detected. The authors suggest that the structural rearrangements of the chromosome disrupted normal segregation of an imprinted region, resulting in UPD and PWS.

Gene structure, DNA methylation, and imprinted expression of the human SNRPN gene
-
Glenn CC, Saitoh S, Jong MTC, Filbrandt MM, Surti U, Driscoll DJ, Nicholls RD: Gene structure, DNA methylation, and imprinted expression of the human SNRPN gene. Am J Hum Genet 1996, 58:335-346. The authors characterize two previously unidentified 5′ exons of the Snrpn gene. They report that this Snrpn gene is expressed in a wide range of somatic tissues and at high, approximately equal levels in all regions of the brain. Their findings are consistent with a key role for DNA methylation in the imprinted inheritance and subsequent gene expression of the human Snrpn gene.

Reports that Snrpn methylation analysis may be useful for the diagnosis of PWS in cultured amniotic fluid or chorionic villus samples and that it will allow, for the first time, prenatal diagnosis for families known to carry imprinting center defects.

Deletion of small nuclear ribonucleoprotein polypeptide N (SNRPN) in Prader-Willi syndrome detected by fluorescence in situ hybridization: Two sibs with the typical phenotype without a cytogenetic deletion in chromosome 15
-
Ishikawa T, Kite T, Wada Y: Deletion of small nuclear ribonucleoprotein polypeptide N (SNRPN) in Prader-Willi syndrome detected by fluorescence in situ hybridization: two sibs with the typical phenotype without a cytogenetic deletion in chromosome 15. Am J Med Genet 1996, 62:350-352. A description of two siblings with typical PWS in whom a small deletion was detected in the PWS critical region by using FISH with a Snrpn gene probe. The authors suggest that this method will permit analysis of persons strongly suspected of having PWS in whom deletions are not detectable by other probes.

Growth hormone treatment in Prader-Willi syndrome: A controlled study
-
Lindgren AC, Hagenas L, Muller J, Blichfeldt S, Lindqvist MA, Ritzen EM: Growth hormone treatment in Prader-Willi syndrome: a controlled study. Horm Ras 1995, 44:61. This is the only controlled study to date that has evaluated the effects of growth hormone therapy in prepubertal children with PWS. Growth hormone test results, including IGF-1 levels, are reported for both children with PWS and obese control subjects. The positive effects of growth hormone on growth velocity, IGF-1 levels, and body mass index are reported. Similar studies of longer duration are still necessary to provide information about long-term benefits and improvement in final adult height in this population of children with PWS.
