The barley HVA1 promoter was obtained by PCR with the use of barley genomic DNA and two primers: 5′-TCCACCGAGATGCCGACGCA-3′ and 5′-GTT-GGAGGCCATGGTCGTCTCACGAT-3′. The HVA1 promoter and the SGFP were fused at the ATG Nco l site. Four clones were selected and tested for stress responses and gave identical results.

Kinase fragments used were as follows: AtCDPK (AK1), amino acids 1 through 413; ATCDPK1, amino acids 1 through 274; ATCDPK1a, amino acids 1 through 274; ATCDPK2, amino acids 1 through 289; ATPKa, amino acids 1 through 284; ATPKb, amino acids 1 through 283; ASK1, amino acids 1 through 265; and ASK2, amino acids 1 through 265. One strand of the sequence for the DHA tag was 5′-CCTTACCCATACGACGTTCCAGACTACGCTGGTT-ACCCATACGACGTTCCAGACTACGCT-3′. All cDNAs were fused to the DHA sequence YPYDVPDYAGYPYDVPDYA (single-letter abbreviations for the amino acid residues are: A, Ala; D, Asp; G, Gly; P. Pro; V, Val; and Y, Tyr) at the Stu I site.
