Cloning and characterization of a cDNA encoding the catalytic subunit of a cAMP-dependent protein kinase from Ancylostoma caninum third-stage infective larvae

3 cDNA library with a catalytic subunit cDNA from C. elegans. The cDNA was found to contain the conserved 5' nematode spliced leader sequence. This paper represents the first reported larval cDNA library, the first signal transduction molecule cloned from a parasitic nematode, and the first report of trans-splicing in hookworms.

3 excretory/secretory products. Western blots using a polyclonal antibody against recombinant ASP 1 were used to characterize the kinetics of ASP 1 release. These experiments indicate that ASP 1 is released prior to the resumption of feeding, suggesting that feeding and ASP 1 secretion are independent events in the activation pathway.

The activity is inhibited by the zinc chelator 1,10-phenanthroline, indicating that the enzyme is a metalloprotease. This is the first report of a proteolytic activity associated with the resumption of development in hookworms.
