This in vitro fusion assay shows that Cdc4Sp, a Sec18p (NSF)-like ATPase, is required to promote ER membrane fusion. This result led the authors to propose that Cdc48p is part of a docking/fusion machinery involved in homotypic fusion that is different from the classical Sec17p (α-SNAP)-Sec18p (NSF) docking/fusion machinery involved in heterotypic fusion. These results, together with results reported in [32••,33••], show that Cdc48p (or its vertebrate homolog, p97) plays a role in homotypic membrane fusion.

Using a morphological in vitro assay, the authors provide evidence that the reassembly of the Golgi stacks after vesiculation with ilimaquinone is sequential and requires at least two steps. The first step is dependent on NSF and α-SNAP, and the second step is dependent upon an NSF-like ATPase, p97. These results support the idea of a role of p97, or its yeast homolog, Cdc48p, in homotypic membrane fusion, as proposed in [31••,33••].

This study showed, using electron microscopy, that Golgi cisternae vesiculated with mitotic extracts regrew with either NSF together with SNAPs or with the NSF-like ATPase p97. However, the morphology of the cisternae rebuilt is different in each condition. This study, together with the findings reported in [31••,32••], raises the possibility that homotypic membrane fusion may implicate p97 or its yeast homolog, Cdc48p.
