Crystal structure of a mixed chain diacylglycerol, 1-stearoyl-3-oleyl-glycerol

of outstanding interest. This structure is of particular interest as it represents the first structure of a saturated/unsaturated mixed-chain lipid. With respect to the V-shaped conformation and molecular packing, it is almost identical to 1,3DBUG [24]. The stearoyl and oleoyl chains pack in separate matrices. Interesting is the neat accommodation of the cis-double bonds into a triclinic chain matrix. The conformational similarity (ap/sc) with the TAG tuning-fork structure with bent α-chain suggests a possible interfacial conformer for 1,3-DAGs. (Note that the molecule in Fig. 1 shows the 1O3SG enantiomer and that atomic co-ordinates and torsion angles have to be inverted to obtain the 1S3O-sn-G enantiomer.)

4/ rotamers sc/, -sc/ and ap/ in solution (65, 31 and 4%) appear to reflect a consistency with the distribution of favoured sc/: -sc/ rotamers in crystal bilayer structures (Fig. 2 and Table 2). The question arises of whether this preference for the sc/ rotamer manifests a favoured minimum energy state of the 1,2-DAG moiety or indicates that 1,2-DAG lipids also in monomeric solution have a tendency for chain stacking.

2D H-NMR conformational study of phosphatidylserine diluted in perdeuterated dodecylphosphocholine micelles. Evidence for a pH-induced conformational transition

of special interest. Chemical shift and nOe data show that phosphatidylserine undergoes a conformational transition that depends on the net charge of the serine head group. In the zwitterionic state (pH 2.6), conformations with layer-perpendicular glycerol backbone (sc/γ/sc and -sc/γ/-sc) and layer-parallel head-group dipoles dominate, whereas with negative net charge (pH 7.5), conformations with layer-parallel glycerol backbone (sc/β/sc and a modified -sc/γ/-sc) and layer-perpendicular head groups are found. The authors also point out the interesting analogy with crystal structures of zwitterionic (DLPE, DMPC) and negatively charged (DLPA-, DMPG-) phospholipids.
