This paper demonstrates that repression blocks translation of oskar mRNA prior to its localization to the posterior pole of the oocyte, when translation is activated. Repression is shown to be mediated by bruno, a protein that binds to multiple BREs in the oskar mRNA 3′ UTR. Failure to bind bruno has lethal consequences, showing that translational control is essential. The relative roles of various genes in mRNA localization and translational control are also defined.

This paper examines the proteins that bind to NREs, the regulatory elements that mediate repression of hunchback mRNA translation. Surprisingly, binding of nanos protein cannot be detected. Proteins that do bind are pumilio and p55. By using a series of double point mutations in the NRE, a strong correlation is established between pumilio binding and NRE function. The correlation for p55, although positive, is not extensive and assignment of a role for p55 in hunchback repression remains tentative.

Translational repression of caudal mRNA by the bicoid homeodomain-containing transcription factor is shown to be responsible for the formation of the caudal protein gradient. Regions in the caudal 3′ UTR bind the bicoid homeodomain in vitro and are required for regulation in vivo. This study goes beyond most others in demonstrating that repression is cap-dependent. Some of the same conclusions are reached in [45], although there are differences in the identification of the 3′ UTR regulatory sequences.
