    • CNTF, a member of the Cytokine family, has similar acute effects in enhancing neurotranemitter release at developing neuromuscular synapses as thai found for neurotrophins. However, unlike neurotrophins, CNTF applied locally at the synapse becomes ineffective when the presynaptic neunte is severed from the soma. A rapid signaling with the cell body appears to be required prior to the potentiation of neurotransmitter secretion

    • Even though BDNF stimulated Ras-GTP formation in sensory and cerebellar granule neurons that had not been exposed to this neurotrophin before, it failed to do so in cultures treated previously for 24 h with BDNF. Concomitant with the functional desensitization, a toss of BDNF binding to its receptor TrkB was observed. In contrast, sensory neurons cultured in the presence of NGF showed no desensitization of the NGF receptor TrkA.

    • Absence of persistent spreading, branching, and adhesion in GAP-43 depleted growth cones
    • This study demonstrates that NGF and IGF-1 elicit distinct behaviors of growth cones of cultured chick sensory neurons. While NGF induced spreading, IGF-1 enhanced branching of growth cones. However, both types of response were abrogated in growth cones that were deficient for GAP-43, a potential modulator of cytoskeletal dynamics and target for phosphorytation induced by growth factors

    • Effects of brain-derived neurotrophlc factor on optic axon branching and remodelling in vivo
    • Using Dil labelling of retinal axons, the authors followed the growth and remodelling of axon branches in living Xenopus tadpoles. Injection of BDNF into ihe optic tectum resulted in a rapid and persistent increase in terminal complexity, whereas other neurotrophins had no effect. Injection of neutralizing antibodies to BDNF produced the opposite effect on axonal branching, suggesting that endogenous BDNF may act to modulate axonal patterning

    • Neurotrophins regulate dendritic growth in developing visual cortex
    • Dendritic length, branching pattern and spine outgrowth of pyramidal neurons in organotypic slice cultures of developing ferret visual cortex were assayed after treatment with various neurotrophins for 36 h. Specific neurotrophins were found to increase the length and complexity of dendrites in a laminar-specific manner
