The existence of hemifusion-a membrane bilayer intermediate between two separate membranes and full fusion-was demonstrated. A solvent-free planar lipid bilayer was exposed lo lipid vesicles containing quenched fluorescent lipids and ionophore. Full fusion was indicated by an increase in planar membrane conductance as ionophore was added to the bilayer. Hemifusion was indicated by a fluorescence flash as lipids escaped from vesicles into the planar membrane under conditions in which full fusion was prevented.

Hemifusion was indicated by a fluorescence flash as lipids escaped from vesicles into the planar membrane under conditions in which full fusion was prevented.

Direct membrane retrieval into large vesicles after exocytosls in sea urchin eggs

Fluorescent dyes of different colors were applied sequentially to fertilized eea urchin eggs. Large (1.5 urn diameter) endosomea that formed were observed to contain only one dye, not a mixture. This, and other evidence, including electron microscopic observations, suggests that the large endosomes formed directly from the plasma membrane, and not by fusion of smaller intermediates.
