Full fusion was indicated by an increase in planar membrane conductance as ionophore was added to the bilayer. Hemifusion was indicated by a fluorescence flash as lipids escaped from vesicles into the planar membrane under conditions in which full fusion was prevented.

    • Direct membrane retrieval into large vesicles after exocytosls in sea urchin eggs
    • Fluorescent dyes of different colors were applied sequentially to fertilized eea urchin eggs. Large (1.5 urn diameter) endosomea that formed were observed to contain only one dye, not a mixture. This, and other evidence, including electron microscopic observations, suggests that the large endosomes formed directly from the plasma membrane, and not by fusion of smaller intermediates.
