    • Real-time measurement of transmitter release from single synaptlc vesicles
    • An elegant and direct measurement of release of single synaptic vesicles of transmitter. The authors suggest that small synaptic vesicles can discharge their entire contents (on average -4000 molecules) with a time course faster than their limit of temporal resolution (60-70 μs). In this paper, Figure 2b and the associated inset show directly that the amount of transmitter released varies from one quantum to the next. This implies that if receptors are not saturated, quantunvto-quanlum variance in transmitter concentration will cause quantum-to-quantum variance in postsynaptic current amplitude.

    • Variation in GABA mini * amplitude Is the consequence of variation in transmitter concentration
    • The authors show that a single release event can generate minis both pre- and postsynaptically ai the same time. The two simultaneous minis are strongly correlated in amplitude. This correlation is interpreted as a dependence of the amplitude of both minis on transmitter concentration, indicating that receptors are not Saturated; the authors then independently verify this conclusion using a pharmacological approach. They estimate that the mean degree of receptor occupancy is less than 50%.

    • Muttlveslcular release from excitatory synapse of cultured hlppocampal neurons
    • In this extension of an earlier study [11], the authors show that the concentration of transmitter in the cleft is sensitive to changes in the probability of release, but the ratio of non-NMDA- to NMDA-receptor-mediated EPSC amplitude is not. This suggests that both receptors experience the same degree of receptor occupancy. Based on the rapid application of glutamate to excised patches, the authors conclude that both receptor types are probably saturated during release.

    • Block of glutamate transporters potentiates ostsynaptlc excitation
    • The authors observe a temperature-dependent increase in miniature and evoked EPSC amplitude in the presence of uptake blockers. A detailed investigation of possible pharmacological artifacts suggests that this effect is genuinely attributable to a decrease in transporter activity. The authors note that the relevant action of the transporters in limiting PSC amplitude need not be uptake per se if the transporters are present in sufficient numbers to buffer rapidly the amount of transmitter released by binding alone. An alternative interpretation of this work has appeared elsewhere [2].

As in the previous report, the authors conclude that most or all variance in mini amplitude is probably attributable to sources of variance intrinsic to each release site In addition, the authors provide evidence that mean mini amplitude at an input is determined by the innervation density on the postsynaptic cell, calling into question the popular assumption that different inputs must have different uniquantal amplitudes.

    • Non-NMDA glutamate sreceptor occupancy and open probability at a rat cerebellar synapse with single and multiple release sites
    • in press. The authors describe two types of synaptic responses isolated by minimal stimulation: the first has a high coefficient of variation (CV), a low amplitude, and a rapid decay; and the second has a low CV, a high amplitude, and a slow decay. This suggests that one type of synapse is not saturated and the other is. The high CV response originates from synapses with a single release site, and the authors estimate receptor occupancy to be -50%. The low CV synapses have a reduced mean postsynaptic current (PSC) amplitude and faster decay when the probability of release is depressed, suggesting that these synapses have multiple release sites, and that 'spill-over' of transmitter from one site to the next is the cause of saturation at those synapses. This indicates that the number of release sites at the synapse is likely to be important in determining the degree of receptor occupancy.

    • The oneveside hypothesis and muftlveslcular release
    • The authors argue that a coordination of multiple release sites within individual synaptic boutons leads to a large number ot multiquantal events in the mini distribution. They make the interesting observation that the distribution of active zones per synaptic bouton matches the number of multiquantal events in the mini distribution implied on the basis of maximum likelihood estimation. If the correlation suggested by this observation existe, the probability of release in a bouton must spontaneously switch between zero and one for all sites in the bouton simultaneously.

    • Saturation of postsynaptlc glutamate receptors after quanta! release of transmitter
    • The authors demonstrate that when two minis are released in rapid succession, the amplitude of the second mini is smaller than that of the first. This implies that a high degree of receptor occupancy takes place during the first mini, supporting receptor saturation in the presence of cyclothiazide. The authors also note the presence of occasional bursts of high-amplitude, low coefficient ol variation (CV) minis, much like the evoked output of the multisite synapses reported elsewhere [50**].
